This dataset contains raw diffraction images collected at SACLA for a paper on SAD phasing of SeMet-labeled proteins: Yamashita et al. (2017) IUCrJ "Experimental phase determination with selenomethionine or mercury-derivatization in serial femtosecond crystallography" doi: 10.1107/S2052252517008557 We encourage you to reprocess this dataset with new programs and ideas. * Datasets This CXIDB entry contains the dataset of ACG (Agrocybe cylindracea galectin). Other datasets described in the paper are deposited in CXIDB #31 and #61. * Geometry MPCCD contains eight panels (512 x 1024 pixels each). In the HDF5 files, eight panels are stacked vertically to give a 512 x 8192 pixel image for a frame. The detector geometry is provided as CrystFEL geometry files. "mpccd-initial.geom" is the original facility provided geometry. "mpccd-optimized.geom" is the refined geometry used for the publication. Unfortunately, because of a problem in metadata handling during data acquisition, the 'adu_per_eV' field of the CrystFEL geometry was inadvertently set to 0. Note that this change affects individual sigI in CrystFEL stream files, but does NOT change the merged sigI, which is based on population standard deviation. * Image processing Image files were filtered and written by Cheetah adapted for SACLA. Details of the data processing pipeline are described in the following paper. "Data processing pipeline for serial femtosecond crystallography at SACLA." Nakane, Takanori, et al. Journal of applied crystallography 49.3 (2016). This format is DIFFERENT from SACLA HDF5 format in the Hg-SIRAS dataset (CXIDB #31). In contrast to the SIRAS dataset, dark current subtraction and relative gain correction have already been applied. Here, one photon corresponds to 10 counts in the file. In other words, the gain is 10. The images are ready for processing by CrystFEL. For example: indexamajig -i files.lst -o out.stream \ -g mpccd-optimized.geom --peaks=zaef \ --threshold=400 --min-gradient=10000 --min-snr=5 --int-radius=4,6,8 -p 3wg3.pdb \ --indexing=dirax,mosflm-cell --integration=rings-nocen-sat \ --fix-bandwidth=5.2e-3 --fix-divergence=4.0e-4 (spot finding parameters should be optimized) * Contact Questions about data processing should be sent to Keitaro Yamashita.
